8 yo, No Quick Fixes for Discipline Problems

Question

Our 8 1/2 year old daughter has not changed any of her undesirable behaviors. These include being irresponsible with schoolwork, unkind to her sister, ruining her clothes/ most other items, eating like an animal at the table and blaming everything on everyone else.
This week I hit a breaking point and banned her to her room everyday after school aside from chores and dinner. She can do homework while in her room and read books for AR test. This punishment does not even phase her. Actually no punishment phases her. Her father and I do not have the best relationship and parenting with him is difficult. He coddled her until she was almost 6. He stopped doing it as much because we adopted again and no one had time to focus all attention on her.
She complains constantly that the kids at school think she is mean and she has no friends. Most days I feel sad for her but don't know what to do.
We have been told she is ADHD but we don't medicate. I actually don't, like John, think ADHD exists. Her biological parents both smoked marijuana while bio mom was pregnant and she may have drank some alcohol. Does this affect the baby? We have always been told no about the marijuana. Both parents were violent and hateful in personality. We have had our daughter since she was born however. So I guess I have two questions, did the drugs affect her brain and how do you punish a child who doesn't care about being punished?
